CAUTIONS
  • Do not use if you have fungal meningitis infections or with ergotamine type drugs, astemizole, lovastatin, midazolam, simvastatin, triazolam, and cisapride.
  • Use with caution if you have impaired liver function; this drug may cause liver toxicity.
  • High doses of ketoconazole may suppress adrenal gland function or the production of testosterone.
DRUG INTERACTIONS
Ketoconazole tablets blocks a specific set of liver enzymes (cytochrome P450 enzymes) that are useful for the metabolism or promote the clearance of other drugs from the body. Ketoconazole interferes with these liver enzymes, and thus, may increase or decrease the clearance of these drugs from the body, often requiring dosage adjustments when they are administered in combination.

Amiodarone, amphetamines, benzodiazepines, beta-blockers, buspirone, busulfan, calcium channel blockers, cyclosporine, sirolimus, tacrolimus, digoxin, docetaxel, HMG-CoA reductase inhibitors (lovastatin, pravastatin, etc.), methylprednisolone, phenytoin, quinolones, trimetrexate, warfarin, sildenafil, tadalafil, vardenafil, and zolpidem. Carbamazepine, didanosine, isoniazid, phenobarbital, phenytoin, rifabutin, and rifampin, protease inhibitors.

Ketoconazole needs gastric acidity in the stomach to be absorbed. The use of antacids, histamine-2 blockers (such as cimetidine, ranitidine, famotidine) and proton pump inhibitors (such as omeprazole, pantoprazole, etc.) may reduce the extent of absorption by lessening the gastric acid production in the stomach. Give these products at least two hours after ketoconazole is taken.

FOOD INTERACTIONS
Avoid alcohol, may cause disulfiram-like reaction (flushing, sweating, increased heart rate, nausea, vomiting). Avoid alcohol ingestion for at least 48 hours after the last dose of ketoconazole.
